Normally, a few amino acids from the C-terminus of the lactamase enzyme are removed through cleavage after it has been translated. The sequence of the original (wild-type) C- terminus can be deduced by comparing it to a mutant in which the reading frame is shifted the insertion or deletion of a nucleotide and the mutant lactamase escapes cleavage.
